ELECTROPORATOR/GENE PULSER BIORAD (B405)

CAPTAIN: Kathy Ruppel - B405

BEFORE USE:

1.) New user MUST be trained by the captain or present users

HOW TO OPERATE:

1.) The usual bacterial transformation settings are: 200ohms, 25 microfarad and 2.5KiloVolts. If you change connections or settings – please return unit to 200ohms and 25 microfarad settings when finished.

2.) The manual is on the shelf next to the unit.

3.) The power switch is behind the main unit – upper left corner – turn on after you have made all connections.

4.) Set Voltage by pressing “set volts” switch on front and then “Raise” or “Lower” keys.

5.) Place cuvette in holder – the tab makes sure that the metal plates will align. Slide it in to make contact with the electrodes.

6.) Press both red buttons “pulse” together at the same time. Display will show charging “Gh9” and then beep when done.

7.) If you do not get a beep and the display flashes – it did not work. Reset unit by turning off power switch and after a few seconds turn it on again.

8.) When done, turn off power and then reset switches and connections.

ADDITIONAL INFORMATION:

- 0.2 mm cuvettes are located in the stock room

- Very High Voltages are used. If you have too much salt in sample, conductivity issues will lead to an arching of the sample. This can cause damage the unit which requires it to be sent back to the manufacturer and large repair costs.

FOR REPAIRS PLEASE SEE KATHY RUPPEL IN B405.
