Week 8, Day 2

Lab work

- Added 2g of sucrose and 0.441g of MS salts to 100ml of sterile water. Added Acetosyringone.

- Spun down agrobacterium cultures grown on 4.8.14

- Resuspended pellets in 10ml of prepared solution

- Left to grow on the bench for 3 hours

- Each construct infiltrated into one nicotiana benthamiana plant on two separate leaves

- Mini preps of level 1 dig-ligs (1,5,9,15,16,17,18,19)

- Quantification of DNA required for next level two dig-ligs
