26.08.2009 1. Isolations of 5,6,8,10 and storages of 2,3,4,5,6,7,8,9,10,11 were made. 2. We tried to make digestions but we couldn't. The following amounts are added to microtubes; 6, water 9.4 µl 18.4µl FD Buffer 2 µl 3 µl Spe1 3 µl EcoR1 3 µl DNA 5.6 µl 5.6 µl 8, water 8.9µl 17.9 µl FD Buffer 2 µl 3 µl Xba1 3 µl Pst1 3 µl DNA 6.1µl 6.1 µl 2nd part, solution 30 µl enzyme 4 µl buffer 4 µl water 2 µl =40µl 3.
