Entry title General Helen Today I looked under my hk032 embryo in the microscope and was excited to start cleaning but realised DiO just didn't penetrate into the hindbrain although I could image it on the nerve stub. I have decided to incubate for another 24 hours to see if that makes a difference. I dyed another stage 20 this morning and loaded it heavily with DiO so see if that makes a difference. I am beginning to wonder if my embryos aren't penetrating properly when I put them in the newer incubator next to the Leica, because that's when DIO seemed to stop working/penetrating into the hindbrain as well. I will see what an extra 24 hours of incubation does to the dye and then consider moving embryos to a different incubator if they still aren't diffusing as well. Fabiana
