Hypocotyl measurement with ImageJ


Needed

  1. ImageJ (from http://rsb.info.nih.gov/ij/)
  2. Working scanner (can be hard)

Scanner Settings

  1. Set to Grayscale.
  2. Set to 600 ppi.
  3. Set to wtb points.
  4. Set to Custom ~130,255.
  5. Set Preferences -> Scan Material -> Positive.
  6. Set Preferences -> More -> Scan Quality -> Quality.
  7. Set Preferences -> More -> Interpolation -> Speed.

Image J

  1. Go to Analyze -> Set Scale.
  2. Over there change:

Distance in Pixels -> 600

Known Distance -> 25.4

Pixel Aspect Ratio -> 1

Unit of Length -> mm
