McClean:YPG

Overview

YPG is a complex medium containing a nonfermentable carbon source (glycerol) that does not support the growth of petite mutants.

Reagents

• Bacto-yeast extract
• Bacto-peptone
• Glycerol
• Bacto Agar
• Distilled water

Protocol

• Weigh out and add ingredients with constant stirring:
  • Bacto-yeast extract (1%) 10g
  • Bacto-peptone (2%) 20g
  • Glycerol (3% [v/v]) 30mL
  • Bacto Agar (2%) 20g
  • Distilled water 970mL
• Autoclave
