BG-11 liquid media
- Cyanobacteria, including Synechocystis PCC 6803, are typically grown in BG-11 media.
- Obtain 50x BG-11 media from Sigma-Aldrich (catalog # C3061-500ML).
- In the biosafety cabinet, mix 20 mL 50x BG-11 media and 980 mL Millipore water.
- Autoclave for 15 minutes at 121C in 500 mL bottles to sterilize.
- Contents of 1x liquid media:
- NaNO3 1.5 g/L
- K2HPO4 0.04 g/L
- MgSO4.7H2O 0.075 g/L
- CaCl2.2H2O 0.036 g/L
- Citric acid 6.0 mg/L
- EDTA 1.0 mg/L
- Na2CO3 0.02 g/L
- Ferric ammonium citrate 6.0 mg/L
- H3BO3 2.86 mg/L
- MnCl2.4H2O 1.81 mg/L
- ZnSO4.7H2O 0.222 mg/L
- Na2MoO4.2H2O 0.39 mg/L
- CuSO4.5H2O 0.079 mg/L
- Co(NO3)2.6H2O 0.0494 mg/L
- Distilled water 1.0 L

BG-11 plates
- For 1 L, or ~2 sleeves of plates, place in a 2L flask:
- 10 mL 1 M TES pH 8.2 (Sigma catalog #)
- 3 g sodium thiosulfate (Sigma, catalog #)
- 15 g Bacto-agar (catalog #)
- 20 mL 50x BG-11 media (Sigma catalog #)
- 980 mL distilled water
- a stirring bar
- Autoclave at 121C for 20 minutes on the liquid cycle.
- Allow to cool, with stirring, until ~50C or cool enough to handle.
