Schumer lab: Changing media

In Xiphophorus cell culture, (as of 11/08/23) media is changed on Monday, Wednesday and Friday. Premade complete media aliquots should be stored in the 4 degree fridge in a 50 mL falcon tube.

Spray the rubbermaid container next to the desktop incubator down with ethanol and place both the media and cells inside the container. Move to the TC hood in the Frasier lab. Spray the inside of the hood and every item thoroughly before placing the cells into the hood.

Carefully turn on the vacuum aspirator in the hood, and place a p1000 pipette tip on the end of the vacuum. Aspirate half of the media from the plates/flasks and replace with equal volume of fresh media.

12 well plates should have a total volume of 2 mL media in each well, while T25 flasks should have a total of 5 mL of media. Return the cells to their original incubator.
