9/6/2016 Flourescence

1. Sample was made in a capped cuvette:

  1. pH 4
    1. Myoglobin: 0.25mM Au, 12.5μM Myoglobin, 3mL total
      1. 225 μL AuCl3
      2. 300 μL 1M HCl
      3. 464 μL Myoglobin
      4. 1871 μL Water


  • Concentration of AuCl3stock was 5.08 mM so concentratio of AuCl3 in final solution was 0.381 mM NOT 0.25mM.
  • Concentration of Myoglobin was 183 uM so concentration of Myoglobin in final solution was 28.304 μM NOT 12.5 μM.


2. The cuvette was heated to 80C and samples were taken every 5 minutes using the fluorescence spectrometer for three hours.


3. Samples for future UV-Vis spectroscopy were made using prepared stock solutions:
