The Daily Grind Describe colony morphology of the new microbes spotted on R2A Mix and ligate 16S fragments from all seeds for cloning (then do the ligation overnight) Cut up the unbacterialized potatoes for propagation Put 10 colonies of ACC deaminase in either pSB1A2 or pDSK-GFPuv in either 1 mL of LGI or 1 mL of LGI plus 100 mM ACC. Include plasmid free E. coli as negative control.
