The Daily Grind

  • Actually doing that PCR today of all the endophytes.
  • Met with Manish and he was OK on the TRFLP using the new nested primers.
  • None of the electroporations with Arabad::GFP worked, not even the positive (E. coli) control. Will have to try again.
  • Setting 20 seeds of B73, Gaspe, Pioneer, and Mixteco to germinate for greenhouse innoculation experiment. They will be innoculated with 100 mL of phosphate buffer with 0D600=0.2 Burkholderia phytofirmans from Mixteco (2A10), Pantoea agglomerans from B73 (1A1), or sterile phosphate buffer
