High Stabilitiy Plasmid Development Abstract Transformation of repC and P1004 ligations Transformations Using the ligations prepared last saturday I transformed competent DH5α cells. I used 5μL of each ligation for 50μL of cells: repC-PCR with B0015 in pSB1AK3 in a 6:4 ratio repC-PCR with B0015 in pSB1AK3 in a 7:4 ratio P1004 with B0014 in pSB1AK3 in a 7:7 ratio P1004 with B0014 in pSB1AK3 in a 9:5 ratio Results Both repC ligations showed colonies possibly containing the plasmid. Plasmid extraction will proceed. No P1004 colonies were observed.
