Objective

The purpose of the present set of experiments was to synthesize gold nanoparticles (in solution and in fibers) and to continue extract proteins.

Description

Nanoparticle Synthesis

In order to make gold nanoparticle fibers BSA and chloroauric acid were combined in a 1:1 ratio (labeled red R), and in order to make nanoparticle fibers the ratio of chloroauric acid to BSA was 1:2 (labeled green G). Two different volumes of the BSA and gold were added for each ratio, thus created 4 test tubes. The green test tubes were left in the oven over night, while the red test tubes were taken out for 50 minutes and allowed to sit at room temperature for 10 min for a total of 3 hours.


  • gA: 250 µL Au (4.17 mM) + 500 µL BSA (30 µM) + 9,250 µL H2O = 1.0425 x 10-4 M Au & 1.5 x 10-6 M BSA
  • gB: 500 µL Au (4.17 mM) + 1000 µL BSA (30 µM) + 8,500 µL H2O = 2.085 x 10-4 M Au & 3 x 10-6 M BSA
  • rA: 500 µL Au (4.17 mM) + 500 µL BSA (30 µM) + 9,000 µL H2O = 2.085 x 10-4 M Au & 1.5 x 10-6 M BSA
  • rB: 1000 µL Au (4.17 mM) + 1000 µL BSA (30 µM) + 8,000 µL H2O = 4.17 x 10-4 M Au & 3 x 10-6 M BSA

Protein Extraction

  • Cells were sonicated (30 sec ON/30 sec OFF 3 times)
  • Solutions were made:
  1. I L 50 mM Tris + 1 M NaCl pH 9, filtered
  2. 1 L 50 mM Tris pH 9, filtered
  3. 4L 50 mM Tris pH 9
  4. 4L 50 mM Tris pH 9
  • cells were placed in dialysis tubes and in 50 mM Tris overnight in a -80 C

Data

  • gA = pinkish purple
  • gB = darker pinkish purple
  • ra = dark purple fibers
  • rb = dark purple fibers
