Objective

Today I'm making samples for electron microscopy and for Doug to take to NIST for dynamic light scattering.

Description

I made two stock solutions for today's work.

HAuCl4

9.3 mg in 10 mL of water = 2.7mM

BSA

9.1 mg in 10 mL of water = 13.6μM

For the 1mL samples the following amounts will be added: 92.6μL HAuCl4, 111μL BSA (for 166 ratio) or 263μL BSA (for 70 ratio), and 796 μL H2O (for 166 ratio) or 644 μL (for 70 ratio).

For the 5mL samples the following amounts will be added: 463μL HAuCl4, 555μL BSA (for 166 ratio) or 1315μL BSA (for 70 ratio), and 3981 μL H2O (for 166 ratio) or 3220 μL (for 70 ratio).

I have 16 samples to make. In all samples the final Au concentration is .25mM. All samples will be held at just under 80°C in an oven or a heat block

  1. Au/BSA concentration ratio 166, made in an eppendorf tube to a final volume of 1mL. This sample is to be left in the heat block for 2 hours. (Sample 1)
  2. Au/BSA concentration ratio 166, made in an eppendorf tube to a final volume of 1mL. This sample is to be left in the heat block for 2 hours. After removing this sample from the heat block it will be immediately placed on ice. (Sample 2)
  3. Au/BSA concentration ratio 166, made in an eppendorf tube to a final volume of 1mL. This sample is to be left in the heat block for 2 hours. After removing this sample from the heat block 0.1 mL of 1.5M tris pH 8.8 will be added. (Sample 3)
  4. Au/BSA concentration ratio 166, made in an eppendorf tube to a final volume of 1mL. This sample is to be left in the heat block for 2 hours. After removing this sample from the heat block it will be placed immediately on ice and 0.1 mL of 1.5M tris pH 8.8 will be added. (Sample 4)
  5. Au/BSA concentration ratio 70, made in an eppendorf tube to a final volume of 1mL. This sample is to be left in the heat block for 2 hours. (Sample 5)
  6. Au/BSA concentration ratio 70, made in an eppendorf tube to a final volume of 1mL. This sample is to be left in the heat block for 2 hours. After removing this sample from the heat block it will be immediately placed on ice. (Sample 6)
  7. Au/BSA concentration ratio 70, made in an eppendorf tube to a final volume of 1mL. This sample is to be left in the heat block for 2 hours. After removing this sample from the heat block 0.1 mL of 1.5M tris pH 8.8 will be added. (Sample 7)
  8. Au/BSA concentration ratio 70, made in an eppendorf tube to a final volume of 1mL. This sample is to be left in the heat block for 2 hours. After removing this sample from the heat block it will be placed immediately on ice and 0.1 mL of 1.5M tris pH 8.8 will be added. (Sample 8)
  9. Au/BSA concentration ratio 166, made in an 13x100mm culture tube (Fisherbrand) to a final volume of 5mL. This sample is to be left in the oven for 4 hours. (Sample 9)
  10. Au/BSA concentration ratio 166, made in an 13x100mm culture tube (Fisherbrand) to a final volume of 5mL. This sample is to be left in the oven for 30 minutes and removed for 15 minutes and placed back in the oven. Repeating the heating/cooling cycle until it has been in the oven for a total of 4 hours. (Sample 10)
  11. Au/BSA concentration ratio 70, made in an 13x100mm culture tube (Fisherbrand) to a final volume of 5mL. This sample is to be left in the oven for 4 hours. (Sample 11)
  12. Au/BSA concentration ratio 70, made in an 13x100mm culture tube (Fisherbrand) to a final volume of 5mL. This sample is to be left in the oven for 4 hours. After removing the test tube it will be placed immediately on ice. (Sample 12)
  13. Au/BSA concentration ratio 70, made in an 13x100mm culture tube (Fisherbrand) to a final volume of 5mL. This sample is to be left in the oven for 4 hours. After removing the test tube 0.5 mL of 1.5M tris pH 8.8 will be added. (Sample 13)
  14. Au/BSA concentration ratio 70, made in an 13x100mm culture tube (Fisherbrand) to a final volume of 5mL. This sample is to be left in the oven for 4 hours. After removing the test tube it will be placed on ice and 0.5 mL of 1.5M tris pH 8.8 will be added. (Sample 14)
  15. Au/BSA concentration ratio 166, made in an 13x100mm culture tube (Fisherbrand) to a final volume of 5mL. This sample is to be left in the oven for 4 hours. This sample is to be left in the oven for 30 minutes and removed to an ice bucket for 15 minutes and placed back in the oven. Repeating the heating/cooling cycle until it has been in the oven for a total of 4 hours. After the full 4 hours it will be placed on ice. (Sample 15)
  16. Au/BSA concentration ratio 166, made in an 13x100mm culture tube (Fisherbrand) to a final volume of 5mL. This sample is to be left in the oven for 4 hours. This sample is to be left in the oven for 30 minutes and removed to an ice bucket for 15 minutes and placed back in the oven. Repeating the heating/cooling cycle until it has been in the oven for a total of 4 hours. After the full 4 hours it will be placed back on ice and 0.5 mL of 1.5M tris pH 8.8 will be added. (Sample 16)

Samples 10, 15, and 16: out at 11:20, in at 11:35, out at 12:05, in at 12:20, out at 12:50, in at 1:05, out at 1:35, in at 1:50, out for good at 2:20 (shorter time than intended)

After the incubation period 1mL of each sample was centrifuged for 10 minutes at 13200 rpm. A UVVis spectrum was taken for each resulting solution. Along with these spectra, I also took a spectrum for: water, 0.25mM HAuCl4, 1.5μM BSA, 3.6μM BSA, 0.25mM HAuCl4 and 1.5μM BSA, and 0.25mM HAuCl4 and 3.6μM BSA.
