Objective

The students will make measurement samples from the stock of malachite green that I made yesterday. We'll measure UV-Vis and fluorescence.

Fluorescence measurement parameters

  1. Excitation wavelength: 555 nm
  2. Emission wavelength start: 565 nm
  3. Emission wavelength end: 700 nm
  4. Excitation slit width: 5.0 nm
  5. Emission slit width: 5.0 nm
  6. Scan speed: 200 nm/min

Notes

Malachite green was not fluorescent, so I made samples of Rhodamine B for fluorescence measurements. The students measured the UV Vis of the malachite green and rhodamine samples and the fluorescence of the rhodamine samples.
