Objective

  • Test the effects of Tris buffer (pH=9.36) and H2O on the formation of a clear purple solution.

Description

1. Four test tubes were prepared:

  • Tube 1 contained 0.1ml of BSA, 0.1ml of HAuCl4, and 0.8ml of H2O.
  • Tube 2 contained 0.1ml of BSA, 0.1ml of HAuCl4, and 0.8ml of Tris buffer (pH 9.36).
  • Tube 3 contained 0.1ml of BSA, 0.05ml of HAuCl4, and 0.85ml of H2O.
  • Tube 4 contained 0.1ml of BSA, 0.05ml of HAuCl4, and 0.85ml of Tris buffer (pH 9.36).

2.The four test tubes were placed in a heatblock set at 80°C for 2 hours.

3. The heatblock containing the four test tubes was turned off to allow the slow cooling of the solutions.

Data

  • For Tube 1, which contained a HAuCl4 to BSA molar ratio of 187, purple fibers formed.
  • For Tube 2, which contained a HAuCl4 to BSA molar ratio of 187, the solution was clear.
  • For Tube 3, which contained a HAuCl4 to BSA molar ratio of 98.4, a purple solution formed. This result was similar to that obtained on September 28th.
  • For Tube 4, which HAuCl4 to BSA molar ratio of 98.4, the solution was clear.
