  • Prepare diluted (1-10µM) solutions of acridine orange and fluorescein
Acridine Orange Calculations
10µM=moles/volume
(10µM)*(0.05L)=5E-7mol
(5E-7mol)*(301.81g/mol)=1.51E04grams of Acridine Orange


Fluorescein Calculations
10µM=moles/volume
(10µM)*(0.05L)=5E-7mol
(5E-7mol)*(332.31g/mol)=1.66E-4grams of Fluorescein



  • Measure Fluorescence of AA stock solutions
Using excitation wavelengths of 308nm, 440nm, and 490nm
Scan speed of 50nm/min
Settings
Excitation wavelength + 10nm
