LB medium (liquid)
- For every 1 L of medium, add to a pyrex bottle or flask:
- 10 g NaCl
- 10 g bacto-tryptone
- 5 g yeast extract
- Add 1 L of deionized water (blue tap of the Millipore machine).
- Autoclave (if using a bottle, do not close the cap completely).
- Let it cool to about 40degC and, when required, add antibiotic near the flame.
- Store in the fridge and always check for contaminations before using.
LB medium (solid)
- For every 1 L of medium, add to a pyrex bottle or flask:
- 10 g NaCl
- 10 g bacto-tryptone
- 5 g yeast extract
- 15 g of agar
- Add 1 L of deionized water (blue tap of the Millipore machine).
- Autoclave (if using a bottle, do not close the cap completely).
- Let it cool to about 50degC and, when required, add antibiotic near the flame.
- Pour approximately 20 ml of medium in Petri dishes (try to avoid bubbles) and let them polymerize at room temperature.
- Cover with foil or parafilm, store in the fridge and always check for contaminations and overdrying before using.
- Use them within 1-2 months.
