I need a category for this page, but can't think of how to label it. Should I use the Ant Sequences Category?

Order Arrived

It is here! Hooray! I will document the information of each oligo below so that anyone can find it and use it in the future. For now I will literally transcribe it by hand, but a .pdf file would be useful too. Other suggestions? (Steve Koch 00:06, 13 November 2009 (EST):transcribing by hand probably easiest way to get pertinent information in...but scanning to PDF and uploading / emailing to kochlab notes is aslo probably good.)

Here is the page I started from. I will do all the resuspensions tomorrow pending Koch's approval.

Top BstXI/SapI

This is for ligating DNA with a specific BstXI overhang to DNA with a specific SapI overhang. We actually have this, but stuff hasn't been working so we ordered new stuff. Info:

  • picomole: 20354
  • micrograms: 257

So I need to resuspend in 203.54ul of 0.1x TE. So weight wise, I will have 257ug in 203ul which is 1.27ug/ul.

Bottom Biotin

This is to complement the top bstxi/sapi oligo above. It has a nick and a biotin so that we can unzip with streptavidin microspheres.
Info:

  • picomole: 30817
  • micrograms: 320

So I need to resuspend in 308.17ul of 0.1x TE. So weight wise, I will have 320ug in 308ul which is 1.04ug/ul.

BstXI adapter top

Similar to top adapter above, but this (in conjunction with BstXI bottom) will ligate 2 DNA fragments with a specific BstXI overhang.

  • micrograms: 246

So I need to resuspend in 194.69ul of 0.1x TE. So weight wise, I will have 246ug in 195ul which is 1.26ug/ul.

BstXI adapter bottom

In conjunction with BstXI adapter top, this oligo (when annealed) will ligate 2 BstXI overhanged DNA fragments together. This is the complement of the above oligo with the same 3' overhang.

So I need to resuspend in 167.7ul of 0.1x TE. So weight wise, I will have 211ug in 168ul which is 1.26ug/ul.

R2342

This is the reverse primer that will allow extension to carry out for an extra 400bp. With this primer I will have 1.5kb PCR products which I will digest at about 1.1kb and gel analyze to determine digestion.

So I need to resuspend in 630.04ul of 0.1x TE. So weight wise, I will have 360ug in 630ul which is 0.57ug/ul.

BstXI adapter bottom biotin

In conjunction with BstXI adapter top, this oligo (when annealed) will ligate 2 BstXI overhanged DNA fragments together. This is the complement of the top adapter oligo of the same name but with the same 3' overhang. This also has a biotin and a nick for unzipping experiments.

So I need to resuspend in 251.56ul of 0.1x TE. So weight wise, I will have 317ug in 252ul which is 1.26ug/ul.
