TAR cloning of natural product gene clusters
- ZB118 subcloning project is underway. The capture vector was mapped with DraI which releases the two targeting sequences for comparison along with a standard fragment map. All clones look fine. JHK and TEG pathway capture vectors also report patterns that are fine (see gel images)
- YAC's isolated from single cells on the cycloheximide selection plates (1:4000 dilution overlay) are in very low concentration. Transformation into EC100's (homebrew) did not yield (#)$. Chemically competent cells also did not yield ()$%. (size restriction for top 10's potentially limiting). e
