Objective

  • Prepare solutions for later round robin experiments

Description

  • Prepared 0.1 M buffer solutions of pH 10, 11, & 12 with Na2HPO4/Na3PO4
  • Used these buffer solutions to prepare Au-AA solutons (in leui of water) for all three AAs (total of 9 solutions)
  • Made a 10 mM HAuCl4 solution and a 100 mM cysteine solution
  • Used these solutions to prepare 10:1, 1:1, & 1:10 Au:AA at each pH (total of 9 solutions)
  • Overnight heated all 18 solutions


  • Prepared Buffer for Bradford Analysis
  • 1L 50mM Tris 50mM NaCl pH 7.5
  • Prepared Buffer for Dialysis
  • 100 mL Glycine Buffer with pH = 3.5


  • Prepared Buffer for SDS-PAGE

3.75mL 0.5M Tris pH 6.8 ---> 6.1042 g Tris in 100mL water ---> 0.504M Tris 15.0mL of 50% glycerol ---> 7.5mL glycerol + 7.5mL water 0.3ml of 1% Bromphenol Blue ---> 0.009g Bromphenol Blue in 1mL water 6.0mL 10% Sodium Dodecyl Sulfate (SDS) ---> 1g SDS in 10mL water H2O to make 30mL total
